Simpler and more efficient experimental process


 

Based on the 3 'end library technology protocol, the 3' end of mRNA was captured using a reverse transcriptome (RT) primer consisting of oligo-dT, UMI and Illumina-compatible 5 'adaptor. During reverse transcription, the oligo-dT acts as a primer to capture the 3 'poly-A tail of the mRNA. The RNA strand in the DNA-RNA hybrid strand is then hydrolyzed using RNase H-specific hydrolase, and the second strand is then synthesized using random primers. Finally, a suitable fragment size is screened and a sequencing-specific library adapter is added to complete the library building experiment. Compared with the traditional transcriptome, it is simplified to only 5 steps, and there is no need for additional poly(A) enrichment, interruption, and 3 'end addition of A.


 


 


 

More targeted analysis process


 

Compared with the traditional transcriptome, the high-throughput transcriptional expression profile based on the 3 'end library technology only sequenced the mRNA sequence near the 3' end, so the data volume requirement is greatly reduced, and only 2Gb is required for the general reference species to reach the sequencing saturation state. High-throughput transcriptional expression profiling follows the quantitative thinking of single-cell transcriptome, and compared with traditional transcriptome, it is comparable in terms of gene quantitative accuracy, functional enrichment consistency and gene detection rate [1].

The new high-throughput transcriptional expression profiling process focuses on gene quantification, gene annotation, differential analysis and enrichment analysis, and can also be associated with LncRNA, miRNA, circleRNA, metabolome and proteome for in-depth study of transcriptional expression regulation mechanisms.

Challenge more difficult samples


 

The traditional transcriptome is to sequence the full-length sequence of mRNA, so the integrity of RNA is required. However, the high-throughput transcription expression profile can be sequenced as long as the structure near the 3 'end of mRNA is complete, so the RNA integrity requirement is not high, and it can be applied to slightly degraded samples, even FFPE samples.



 

By setting different amounts of RNA for samples with different degrees of degradation, it can be found that slightly degraded samples (3<RIN<7) can be found that by increasing the amount of RNA, the final number of genes detected can be comparable to that of normal samples.
